I know a lot of us have had this problem, I myself have had it a couple of times but previously it was always body shop related, and I was able to fix it using the 50/50 method. This time I don't know what to do.
The problem WAS with my BS, but I used the 50/50 method and found three problem files that were in the saved sims folder, NOT the downloads folder. Body Shop worked fine after removing them, but my game itself isn't working. Everything goes fine until I get to the neighborhood screen, and when I go to load one of them, I get the popup. I'm seriously lost as to what to do next.
Would reinstalling help? And if so, what files/folders do I need to keep if I want to keep my sims and downloads? I'm pretty sure that the problem must be some recent downloads, because I haven't made any new sims in a long time, but Body Shop is working so I can't figure out which downloads are responsible. I'm a little afraid to do the 50/50 method again, because I'm not sure how the game would run if I were to remove downloads that sims are currently using. Would that crash the game or would that just replace the custom items with maxis ones?
Any help would be greatly appreciated, I'd hate to lose my sims. I've grown so attached to them.

if the objects you delete are cloned from maxis objects they will be replaced with maxis items. however there is no risk in removing things from your download folder. rather the opposite.

Regardless of what you remove, aslong as you don't save the game the effects aren't permanent. The 50-50 method is time consuming but is most likely the solution. First, remove your Downloads folder as a whole and place it somewhere like your Desktop, run the game and if the error persists you know it's not custom content conflicting. Use this tutorial (http://www.modthesims2.com/showthread.php?t=139718) to find the problem folder.
If it doesn't, then the 50-50 method should solve it. But let us know how it goes :D and I recommend backing up important folders in a separate folder, follow this tutorial (http://www.modthesims2.com/showthread.php?t=138900)

Thank you for the helpful links. According to that first guide, I was right and it isn't my downloads folder or apparently any other particular folder for that matter, which means it must be the game files themselves. I tried deleting everything I could safely delete, and I still get the dreaded error. Unfortunately it looks like I'm going to have to reinstall. :[ I have one question before I do, though. Should I uninstall ALL of my expansions, or will removing the latest one do the trick? I have two EPs, and a slow computer so I'd like to save some time if at all possible haha.

When there are problems with the game files, it's safer to reinstall everything since you can't be sure what particular files aren't working correctly. But before you uninstall, make sure that you backup your entire My Docs/EA Games/Sims2 somewhere safe including compressing it to a .zip or .rar file since occasionally even if the folder is moved it will get deleted upon uninstall. Follow the instructions in this faq to do a clean install: http://www.modthesims2.com/article.php?t=139059

Well, I followed everything in the FAQ to uninstall, and now I can't REINSTALL. Apparently disk 1 of the base game is too damaged or something, because it starts installing, gets so far, and then stops. The disk spins like mad in the drive but nothing ever happens. I seriously can't believe my luck, haha.
So I guess the only solution now is waiting however many months/weeks until I can buy a new copy. Does anyone know if they still sell the base game by itself, without extra EPs or stuff packs? Ordering online isn't an option.

Try getting a replacement disc. EA sells them for like 10 dollar. Info on that on the official sims2 site.
